Detection of PCB on gauze wipes

RANGE OF DETECTION: Low range: 5 to 100 mg per wipe, High range: 25 to 500 mg per wipe

MATERIALS:
PCB RaPID AssayÒ Kit and Sample Diluent. RaPID Prepä PCB Sample Extraction Kit. Reagents: hexane (pesticide grade). Equipment: 3 inch x 3 inch cotton gauze pads (packaged individually as sterile gauze pads); 5 mL serological pipets or equivalent dispenser for hexane; forceps; a 1.0 mL repeating pipet and a precision pipet capable of delivering 250 mL (required for use with the PCB RaPID Assay Kit); 1.5 mL capacity glass test tubes or vials (available from Ohmicron, Part# A00192); protective gloves.

WIPE PROCEDURE:
Caution: Wear protective gloves for this procedure. Avoid breathing hexane fumes. Saturate a wipe with at least 5 mL of hexane. Wipe a 10 cm x 10 cm area (100 cm2) with the saturated wipe. Change gloves between wipes to avoid cross-contamination. Place wipe on a clean surface, such as aluminum foil, and allow it to air dry (at least 10 minutes). Using the forceps, transfer the dry wipe to a bottle of RaPID Prep PCB Extraction Solution (20 mL). Shake vigorously for about 20 seconds. Allow any particulates to settle before proceeding.

DILUTION PROCEDURE:
A. For detection of PCB in the low range: Using the pipet provided, transfer 25 mL of the extract directly into a vial of RaPID Prep PCB Extract Diluent (25 mL). Mix by inverting several times. This mixture is now measured as the 'sample' according to the package insert of the PCB RaPID Assay Kit.
B. For detection of PCB in the high range: Dispense 1.0 mL of the PCB Sample Diluent into a test tube or vial (one for each sample). Follow instructions in step A. Transfer a 250 mL aliquot from the PCB Extract Diluent vial to the 1.0 mL Sample Diluent. This mixture is now measured as the 'sample' according to the package insert of the PCB RaPID Assay Kit.
