Detection of PCB on gauze wipes using the rapid assay test system

RANGE OF DETECTION: Low range: 5 to 100 mg per wipe. High range: 25 to 500 mg per wipe

MATERIALS:
PCB RaPID AssayÒ Test Kit (A00133) and Sample Diluent (A00136). PCB Wipe Sample Extraction Kit (A00137WA). Reagents: Methanol (ACS Reagent grade). Equipment: 3 inch x 3 inch cotton gauze pads (packaged individually as sterile gauze pads); forceps; 1.5 mL capacity glass test tubes or vials; protective gloves. Rapid Assay Field System (along with appropriate pipet tips for the repeater pipette and adjustable volume pipet). (Part #6997010)

WIPE PROCEDURE:
Caution: Wear protective gloves for this procedure.
Saturate a wipe with at least 5 mL of methanol (from a solvent ampule in the Rapid Assay PCB Sample Extraction Kit). Add remaining methanol to an extraction jar. Wipe a 10 cm x 10 cm area (100 cm2) with the saturated wipe. Change gloves between wipes to avoid cross-contamination.
Using the forceps, transfer the wipe to the extraction jar. Continue to follow wipe procedure in the Extraction Kit User’s Guide.

DILUTION PROCEDURE:
A. For detection of PCB in the low range: Using the pipet provided, transfer 25 mL of the extract directly into a vial of RaPID Prep PCB Extract Diluent (25 mL). Mix by inverting several times. This mixture is now measured as the 'sample' according to the package insert of the PCB RaPID Assay Kit.
B. For detection of PCB in the high range: Dispense 1.0 mL of the PCB Sample Diluent into a test tube or vial (one for each sample). Follow instructions in step A. Transfer a 250 mL aliquot from the PCB Extract Diluent vial to the 1.0 mL Sample Diluent. This mixture is now measured as the 'sample' according to the package insert of the PCB RaPID Assay Kit.
