Many qPCR users rely on reaction mixes formulated with ROX reference fluorophore. When an instrument is described as “no-ROX”, a common concern is whether ROX-containing mixes must be avoided or whether quantification could be affected. Our new technical note for the FastGene® qFYR explains this clearly. While ROX is not required for reliable quantification on the qFYR, it is an open system compatible with no ROX, low ROX, and high ROX master mixes. We observed equivalent Cq values and comparable standard curve metrics across ROX conditions.
