Synthego - CRISPR Nuclease

Navigating the SpCas9 IP landscape for a CRISPR therapeutic is challenging, often resulting in delays in development. To accelerate the next generation of CRISPR-based therapeutic development by making CRISPR more accessible, we introduce our novel high-fidelity nuclease, hfCas12Max. Engineered from the CRISPR Cas12i nuclease to enable broad PAM sequence recognition and high fidelity (high editing efficiency and low off-target) across a wide range of cell types, hfCas12Max is the newest nuclease member of the type V Cas12 system. Due to hfCas12Max nuclease's smaller size (1080 amino acids) compared to SpCas9 nuclease (1368 amino acids) and LbCas12a nuclease (1228 amino acids), it is easier to package inside lipid nanoparticles (LNPs) or adeno-associated viruses (AAVs).

Since its discovery in the Nobel Prize-winning publication, Cas9 nuclease has been used repeatedly in various applications spanning protein visualization studies, the generation of disease models, agriculture improvements, and most notably, in the cell and gene therapy space making it the gold standard in CRISPR gene editing. Of the different Cas9 nuclease variants, the most widely known is Streptococcus pyogenes (SpCas9). To maximize its efficacy, researchers and scientists know that complexing SpCas9 nuclease with synthetic sgRNA enables them to achieve desirably high editing efficiencies needed to be successful in their CRISPR experiments and therapeutic development. However, not all SpCas9 nuclease variants sourced from different companies provide the high efficiency needed to be successful at gene editing.

T7 RNA Polymerase is an enzyme known for its precision in transcribing RNA from DNA sequences governed by T7 phage promoters. With a molecular weight of around 99 kDa, it is indispensable for synthesizing RNA in vitro, thanks to its unmatched specificity for T7 promoters, ensuring efficient and controlled transcription.
