M I C R O P L A T E E V A P O R A T O R SMicroplate evaporatorsPorvair evaporators are designed to removethe traditional laboratory ‘bottleneck’ ofsolvent evaporation from microplates prior toanalysis or reconstitution in storage buffer.These evaporators give significant throughputadvantages to laboratories looking to optimisemicroplate sample preparation productivity.Faster than centrifugal evaporation,significant increases in sample throughput areachieved through advanced evaporator headtechnology and an innovative manifolddesign, which directly injects heated nitrogeninto each individual well of the microplatesimultaneously. The evaporators have beendesigned to be simple to install, operate andmaintain. Installation requires only connectionto a gas supply or cylinder and mainselectricity. Safety of operation is ensured asthe CE marked compact units fit into all fumecupboards. Not suitable for high boilingsolvents such as DMSO and water.Both MiniVap and UltraVap may be operatedwith a supply of clean, dry compressed air inplace of nitrogen, if the chemistry allows. Warm gas is blown down intothe wells of the microplate, just above theliquid level. The effect is to speed up solventevaporation by providing more energy for thelatent heat of evaporation. This enables asteady-state equilibrium to be reached morequickly, leading to shorter drying times.Nitrogen blow-down has been shown to bethe easiest way to automate the frequentbottlenecks caused by the requirement toremove solvent from samples that need to beconcentrated, dried or reconstituted in a moresuitable solvent.Flexible programmingThe Ultravap RC has been designed to allowrobots with extended gripper arms to placeand remove microplates directly onto theevaporation table. In comparison withprevious models, the Ultravap RC is22mm shorter at the front end, allowingbetter access for the gripper arms. Theevaporation table is able to rise under thecontrol of a stepper motor as the dryingprocess proceeds. This can be programmed ata suitable rate for each solvent type beingevaporated. In addition, gas temperature,pressure and flow rate can all be programmedindividually and stored in one of ninemultistep programmes on the UltravapRC. Each programme allows the tableto rise in two distinct ramped phases,so that a fast initial drying period canbe followed by a gentler final dryingphase.With the addition of the optionalclear acrylic side sheets and the 4inch fume duct adaptor, the newUltravap RC unit lends itself to fullintegration with Tecan and Hamilton roboticliquid handlers, where it is usually located onthe right-hand side of the deck. Controlcommands are sent directly from the robotcontroller to the Ultravap. These standardcommands are listed in the manual, but mostrobot manufacturers have drivers available tocontrol the Ultravap, making integration aseamless process.Solvent compatibilityWith a choice of 96 straight or 96 spiral needles, plus 24 and384 straight needles, the sphere of application for UltravapRC is huge. Most common chromatography solvents can beevaporated with ease, such as dichloromethane, methanol,acetonitrile and hexane. The nitrogen blow-down method isnot, however, suitable for high-boiling point solvents such asDMF, DMSO and water. It is also unable to be used with acidsor acid chlorides, as special corrosion-resistant systems areneeded for this work. The choice of straight or spiral needlesallows the user to choose between faster dry down (spiral)and better final drying in V-well plates (straight). The spiralscause a vortex to form in the solvent, increasing the surfacearea and thus speeding up the rate of evaporation.
