The successful retention and quantification of polar analytes and metabolites continues to be an exceptional chromatographic challenge for analytical scientists.  Hydrophilic interaction chromatography (HILIC) can be used as a chromatographic technique to successfully improve the retention of very polar species while improving sensitivity in mass spectrometry, shortening sample handling steps and provide orthogonal selectivity compared to RPLC.  Due to the complex, multi-modal retention mechanisms present in HILIC, predicting retention behavior to develop HILIC separations can be rather challenging. Further difficulties arise due to the altered mobile phase pH and analyte pKa values in highly organic HILIC solvents.
