  1. Prepare thin blood films and fix in Reagent 1 for 3 min
  2. Dilute one volume of Reagent 2 with four volumes of distilled water, flood the slide and allow the stain to act for 5-20 min.
  3. Wash and differentiate with dist.water, controlling the degree of differentiate microscopically.
  4. Drain and dry in the air at room temperature.
  • Nuclei of leucocytes : Reddish-purple.
  • Eosinophilic granules : Red to orange.
  • Basophilic granules : Blue.
  • Lymphocytes : Dark purple.
