Prior to use, prepare a 1 mg/mL working stock dilution in CUT&RUN Antibody Buffer (20 mM HEPES pH 7.5, 150 mM NaCl, 0.5 mM Spermidine, 0.01% Digitonin, 2 mM EDTA). Store working stock at 4°C for 1 month. Use 0.5 µg of the working stock for each CUT&RUN assay. A typical yield for a CUT&RUN assay using 500,000 K562 cells is 2-5 ng DNA.
