Sartorius - In-line Alkaline Lysis System for pDNA Isolation
The In-line Alkaline Lysis System offered by Sartorius is a state-of-the-art, automated solution designed for efficient process development and pilot-scale operations. It features a closed single-use loop system capable of processing up to 500 liters of resuspended bacterial cells per workday. Integrating advanced mixing controls allows for continuous operation, enhancing the isolation of plasmid DNA by minimizing contamination from host cell genomic DNA and RNA. The system comprises the Lysis Reactor and the Alkalizator Tower, which collectively streamline the lysis process. The Lysis Reactor, a single-use component, is designed for ease of installation and operation in compact spaces, providing an effective alternative to traditional batch lysis methods. The Tower features user-friendly software, four peristaltic pumps, and ultrasonic flow sensors for precise control, alongside essential safety precautions like non-contact pressure sensors to ensure reliable performance.
Our In-line alkaline lysis system is designed to increase control and efficiency of pDNA isolation from bacterial cells, while decreasing unwanted impurities such as host cell genomic DNA and RNA. The Lysis Reactor is a single use component with simple installation, requires little countertop space and provides a clean and simple alternative to larger batch lysis processes allowing user to use it as a plug and play combination.
Components of the In-line Lysis System
The Tower
The Tower serves as the controller unit in the system with user friendly software. It contains four peristaltic pumps that are precisely controlled via ultrasonic flow sensors, ensuring tight control over process conditions. The System’s Tower also contains all safety precautions, such as non-contact pressure sensors, to allow for safe process execution.
The Lysis Reactor
The Lysis Reactor is a single-use component to use in combination with the Alkalizator Tower. Together they form fully automated in-line lysis system with a closed single-use loop for process development (PD) up to pilot scale. The Lysis Reactor consists of the main reactor body with a fixed volume of 1000 mL or 5000 mL, 5 inlet paths (4 solutions and one air) and an outlet path.
